Pyruvate and glyceraldehyde-3-phosphate condense via DXS to form the universal Cβ‚… isoprene building blocks IPP and DMAPP, the entry point for all terpenoids.

GPP formation

Geranyl diphosphate synthase (GPPS) catalyses the head-to-tail condensation of IPP and DMAPP to form GPP, the direct monoterpene precursor fed into cannabinoid assembly.

Polyketide synthesis

Hexanoyl-CoA is extended by olivetol synthase (OLS) and cyclised by olivetolic acid cyclase (OAC) to produce olivetolic acid, the phenolic moiety of all cannabinoids.
